Comparative assessment of dermal wound healing potentials of various Trifolium L. extracts and determination of their isoflavone contents as potential active ingredients

- Ethnopharmacological relevance:Trifolium species areusedinTurkishfolkmedicineasawoundhealing
agent, expectorant,antiseptic,sedativeandtoalleviatepaininrheumatism.Inthepresentstudy,the
aqueousmethanolicextracts(80%)of13 Trifolium species (Trifoliumambigum, Trifoliumarvense var.
arvense, Trifoliumcampestre, Trifoliumcanescens, Trifoliumhybridum var. anatolicum, Trifoliumhybridum
var. hybridum, Trifoliumpannonicum, Trifoliumpratense var. pratense, Trifoliumpurpureum var. purpureum,
Trifoliumrepens var. repens, Trifoliumresupinatum var. microcephalum, Trifoliumspadiceum and Trifolium
trichocephalum) collectedfromdifferentregionsofAnatoliawereevaluatedfortheir in vivo wound
healing effects.
Materials andmethods:Invivo woundhealingactivitiesoftheplantaqueousmethanolicextractswere
evaluatedbylinearincisionandcircularexcisionwoundmodelssubsequenttohistopathological
analysis.Activeconstituentsweredeterminedbyavalidatedhighperformanceliquidchromatographic
method. Precisionofthemethodwasperformedbytheevaluationofintra-dayandinter-dayvariations
of theeachstandardatlimitsofquantification (LOQ)levels.
Results: The aqueousmethanolicextractsof Trifoliumcanescens and Trifoliumpretense var. pratense
possessed betterwoundhealingactivitycomparedtotheotherextractsandcontrolgroups.Theanimal
groups treatedwiththe Trifoliumcanescens extractdemonstratedincreasedcontraction(48.96%)on
excisionandasignificant increaseinwoundtensilestrength(35.6%)onincisionmodels.Themain
compounds weredetectedasgenisteinandbiochaninAfor Trifoliumcanescens.
Conclusion: The resultsofthepresentstudyrevealedthewoundhealingpotentialof Trifoliumcanescens.
This mightbeduetothecombinedeffectoftheisoflavonesgenistein,formononetin,daidzein,and
biochanin Apresentintheextract.
